5-FU and Calcipotriol (Topical)

A combination topical therapy (5-fluorouracil plus the vitamin D analog calcipotriol) used as a field treatment for actinic keratoses, sun-induced precancerous skin lesions. Calcipotriol induces the alarmin TSLP, priming a CD4+ T-cell immune response that, with 5-FU, clears precancerous keratinocytes; randomized-trial evidence supports efficacy for actinic keratosis. Its relevance to skin healthspan is as a precancer field therapy that may also improve the appearance of photoaged skin.

Protocol

• Apply a topical cream containing 0.005% calcipotriol and 5% 5-fluorouracil twice daily for 2-4 days.

Flags

• Causes significant skin irritation, redness, and peeling as part of its mechanism of action.
